N-Glycosylation Profiles of the Green Microalgae Chlorella Zofingiensis

Abstract
 Nowadays, the use of microalgae species as raw materials in biopharmaceutical production is on agenda. The reason behind this idea that are cell factories able to efficiently utilize carbon dioxide for numerous biologically active compounds. However, there several problems remain be solved recombinant protein from microalgaes. One critical requirements produce a bio-compatible N-glycosylation profile secreted proteins. knowledge about glycosylation machinery and N-glycan profiles spices quite limited. In study, it was aimed characterize green microalgae, Chlorella zofingiensis. To achieve this, photoautotrophically grown zofingiensis extracts including (glyco-)proteins were enzymatically deglycosylated labelled with 2-aminobenzoic acid tag. Released N-glycans purified HILIC-based approach analyzed by MALDI-TOF(/TOF)-MS. results showed C. included oligomannosidic type patterns. addition, MALDI-MS revealed most phosphorylated.
 
 Öz
 Günümüzde, mikroalg türlerinin biyofarmasötik üretiminde hammadde olarak kullanımı konusu tartışılmaktadır. .Bu fikrin altında yatan temel neden mikro alglerin, biyolojik aktif çok sayıda bileşenin üretimi için karbon dioksiti etkin bir şekilde kullanabilen hücre fabrikaları olmasıdır. Bununla birlikte, mikroalglerden rekombinant kullanılması çözülmesi gereken birkaç sorun mevcuttur. Kritik gereksinimlerden birisi salgılanan proteinlerden biyo-uyumlu N-glikozilasyon üretmektir. Ancak glikozilasyon makineleri ve N-glikan profilleri hakkındaki bilgiler oldukça sınırlıdır. Bu çalışmada, yeşil türü olan ‘in profilinin karakterize edilmesi amaçlandı. amaçla (gliko-)proteinleri içeren fotoototrofik yetiştirilmiş ekstreleri enzimatik deglikozile edildi 2-aminobenzoik asit etiketi ile etiketlendi. Serbest hale getirilen N-glikanlar HILIC bazlı yaklaşımla saflaştırıldı MALDI-TOF (/TOF)-MS analiz edildi. Sonuçlar zofingiensis'in oligomannozidik türde modellerini içerdiğini göstermiştir. Ek olarak, tarafından N-glikosilasyon profilleri, oligomannosidik N-glikanların neredeyse yarısının fosforilenmiş olduğunu ortaya koymuştur.
